Satraplatin + prednisone is an investigational oral combination therapy primarily evaluated for the treatment of hormone-refractory prostate cancer (HRPC), also known as castration-resistant prostate cancer (CRPC). Satraplatin is a third-generation oral platinum(IV) compound that functions as a DNA-damaging agent—when cellular uptake occurs, it forms DNA adducts and cross-links, disrupting DNA synthesis and replication, ultimately inducing apoptosis. Prednisone is a corticosteroid with anti-inflammatory and immunosuppressive effects, and in the context of prostate cancer, it provides palliative benefits and may enhance chemotherapeutic efficacy. Clinical trials have demonstrated that the combination results in a statistically significant improvement in PSA responses and progression-free survival versus prednisone alone in patients with HRPC; overall survival, however, was not significantly extended, leading to discontinuation of large-scale development. Toxicity was generally mild. Both drugs are administered orally[1][2][3][4][6].
