Overview

Sauchinone is a bioactive lignan compound isolated from the medicinal plant *Saururus chinensis*. It exhibits a range of pharmacological effects including anti-inflammatory, antioxidant, anti-obesity, anti-hyperglycemic, hepatoprotective, and neuroprotective activities. Mechanistically, sauchinone exerts significant anti-inflammatory actions by inducing heme oxygenase-1 via ERK signaling, suppressing mediators such as COX-2 and iNOS in macrophages, and modulating the NF-κB pathway. In liver cells, it activates Nrf2, thereby promoting the expression of cytoprotective genes and protecting against oxidative and chemical injury. In cancer models, sauchinone inhibits cell proliferation, particularly in breast cancer, by upregulating miR-148a-3p, thus downregulating HER-2, and in colorectal cancer, by downregulating PD-L1 and MMP2. It also helps regulate cholesterol homeostasis in the liver, reducing hepatic steatosis and LDL cholesterol by downregulating PCSK9 via SREBP-2. Other reported effects include renoprotection, gastrointestinal benefits (e.g., in ulcerative colitis), anxiolytic effects mediated via nitric oxide signaling, and potential benefits in metabolic and inflammatory diseases. While preclinical studies are promising, clinical efficacy in humans remains to be established[1][2][3][4][6][7][9][10].
