Overview

SB 11285 is a second-generation small molecule agonist of the Stimulator of Interferon Genes (STING) pathway, developed for intravenous administration. It activates the STING pathway in immune cells within the tumor microenvironment, leading to downstream activation of TANK-binding kinase 1 (TBK1), nuclear factor-kappa B (NF-kB), and interferon regulatory factor 3 (IRF3). This results in induction of both innate and adaptive immunity, including activation and priming of cytotoxic CD8+ T cells and natural killer (NK) cells for anti-tumor responses. Preclinical studies have shown that SB 11285 reduces tumor volumes as monotherapy or in combination with checkpoint inhibitors such as anti-PD-1 or anti-CTLA-4 antibodies. The drug is systemically administered, enabling access to hard-to-reach tumors and facilitating migration of activated immune cells into tumor sites. It is being investigated primarily for advanced solid tumors, including melanoma and head and neck squamous cell carcinoma[1][2][3][4][5][6][7][8].
