SB-271046 is a potent, selective, and orally active small molecule antagonist of the serotonin 5-HT6 receptor. It was one of the first selective 5-HT6 antagonists, discovered through high-throughput screening at SmithKline Beecham. It competitively blocks the 5-HT6 receptor, significantly increases seizure threshold in animal models, selectively enhances extracellular glutamate levels in brain regions linked to cognition, and shows cognition-enhancing and anti-amnesic properties in preclinical models. It is primarily used as a research tool to explore the central function of 5-HT6 receptors and potential therapeutic strategies for cognitive dysfunction and epilepsy. Despite oral activity and selectivity, it exhibits poor blood-brain barrier penetration in vivo and is not clinically approved for any indication[1][7][13][14].
