SB010 is an inhaled DNAzyme-based GATA-3 antagonist developed for the treatment of Th2-driven inflammatory diseases such as asthma and chronic obstructive pulmonary disease (COPD). It belongs to a new class of antisense oligonucleotide therapeutics known as 10-23 DNAzymes. The drug specifically targets and cleaves GATA-3 mRNA, thereby inhibiting the expression of this master transcription factor that regulates Th2-mediated inflammation. By reducing GATA-3 activity, SB010 decreases the production of key cytokines including interleukin (IL)-4, IL-5, and IL-13—central mediators in allergic airway inflammation. Clinical studies have shown that inhaled SB010 can attenuate bronchospasm after allergen challenge in asthma patients and reduce sputum eosinophils in both asthma and COPD patients. The drug has demonstrated a favorable safety profile with no serious adverse events reported in clinical trials[1][4][5][6][10].
