SB157 is a novel heterocyclic pyrimidine small molecule that functions as a tubulin inhibitor by targeting the colchicine binding site. Developed at the University of Tennessee Health Science Center, SB157 inhibits tubulin polymerization, leading to cell cycle arrest and apoptosis in cancer cells. It also exhibits vascular disrupting activity, causing rapid disruption of established tumor vasculature. In preclinical models, SB157 has demonstrated significant efficacy against melanoma and paclitaxel-resistant prostate cancer, suggesting its potential to overcome common mechanisms of multidrug resistance associated with taxane-based therapies.
