Overview

SB204 is a first-in-class topical nitric oxide-releasing drug candidate developed by Novan, Inc. for the treatment of acne vulgaris. The product utilizes Novan's proprietary Nitricil technology, which consists of nitric oxide-releasing silica nanoparticles. Upon application, the gel releases nitric oxide, which exerts a multi-pronged therapeutic effect: it acts as an antimicrobial agent against *Cutibacterium acnes*, reduces the production of sebum by sebocytes, and inhibits the NLRP3 inflammasome to provide anti-inflammatory benefits. In 2017, Novan reported results from two pivotal Phase 3 clinical trials (NI-AC301 and NI-AC302); while the drug demonstrated statistical significance in one trial, it failed to meet primary endpoints in the second, leading to the discontinuation of its development for acne.
