SB225002 is a potent, selective, and non-peptide small molecule antagonist of the chemokine receptor CXCR2 (also known as IL-8RB). It inhibits interleukin-8 (IL-8) binding to CXCR2 with an IC50 of approximately 22 nM and displays over 150-fold selectivity for CXCR2 over other related receptors such as CXCR1. SB225002 blocks IL-8-induced neutrophil chemotaxis and margination both in vitro and in vivo. It has been shown to reduce inflammatory responses in animal models of colitis by decreasing neutrophil influx and pro-inflammatory cytokines. Additionally, it demonstrates antitumor activity by inhibiting cell proliferation and tumor growth in preclinical cancer models. The compound is widely used as a research tool for studying the role of CXCR2 signaling in inflammation, immune response, cancer biology, and radiosensitization[1][3][5][7][10].
