SB414

SB414 is an investigational nitric oxide-releasing topical cream developed by Novan, Inc. (and later acquired by Ligand Pharmaceuticals) for the treatment of inflammatory skin conditions, including plaque psoriasis and atopic dermatitis. The product is a two-component system consisting of a stable ointment containing the active pharmaceutical ingredient NVN1000 (a nitric oxide-releasing macromolecular polysiloxane) and a hydrogel, which are mixed prior to application. SB414 works by delivering nitric oxide to the skin, which subsequently downregulates the expression of pro-inflammatory cytokines such as Interleukin-17A (IL-17A) and Interleukin-17F (IL-17F), key drivers of psoriatic lesions. The drug has been evaluated in Phase 1b clinical trials for mild-to-moderate chronic plaque psoriasis and Phase 2 trials for atopic dermatitis.
