SBI-100 is a proprietary amino acid ester prodrug of delta-9-tetrahydrocannabinol (THC), developed as a topical ocular formulation for the treatment of glaucoma and ocular hypertension. It is formulated as an ophthalmic nanoemulsion designed to enhance corneal penetration and bioavailability. After administration, the prodrug crosses the corneal membrane and is converted into active THC in ocular tissues, where it acts as an agonist at cannabinoid receptor type 1 (CB1). Activation of CB1 receptors in key ocular tissues can lower intraocular pressure (IOP), addressing a major therapeutic need in glaucoma management. Preclinical studies demonstrated IOP-lowering efficacy comparable to standard-of-care treatments without psychotropic side effects. Clinical trials showed that SBI-100 was safe, well-tolerated, and effective at reducing IOP in healthy volunteers with elevated baseline IOP.
