SBS-518 is a first-in-class, orally administered small molecule in development for the treatment of stimulant use disorder (StUD), including cocaine and methamphetamine use disorders. It acts as a dual antagonist of sigma receptors (both S1 and S2 subtypes) and an inhibitor of the dopamine active transporter (DAT). By targeting both sigma receptors and DAT, SBS-518 blocks the stimulant-induced rise in extracellular dopamine, thereby reducing drug-seeking behavior without producing self-rewarding effects. The compound was developed by researchers at the University of Florida College of Pharmacy, led by Dr. Christopher R. McCurdy, in collaboration with Sparian Biosciences. Preclinical studies have demonstrated efficacy in animal models for blocking cocaine and methamphetamine self-administration while maintaining normal behaviors such as food intake[3][5][6][8].
