SBT6050 is an investigational Immuno-Targeted Antigen Conjugate (ImmunoTAC) developed by Silverback Therapeutics. It consists of a humanized monoclonal antibody that binds to the HER2 sub-domain II (the pertuzumab epitope) conjugated to zuvotolimod, a potent and selective small molecule Toll-like receptor 8 (TLR8) agonist. The therapeutic is designed to systemically deliver the TLR8 agonist to HER2-expressing tumors, where it locally activates myeloid cells—such as macrophages, monocytes, and dendritic cells—within the tumor microenvironment. This activation is intended to trigger both innate and adaptive immune responses against the cancer cells. SBT6050 was primarily developed for the treatment of advanced HER2-positive or HER2-expressing solid tumors, including breast, gastric, colorectal, and non-small cell lung cancers. However, development of the program was discontinued in 2022 following interim clinical data.
