SBT6050 + pembrolizumab is an investigational combination therapy being evaluated for the treatment of advanced or metastatic HER2-expressing or amplified solid tumors. SBT6050 is a novel antibody–drug conjugate that consists of a HER2-directed antibody (binding the pertuzumab domain) conjugated to a Toll-like receptor 8 (TLR8) agonist through a chemical linker. This design delivers targeted innate immune activation to the tumor microenvironment: SBT6050 directly activates myeloid cells (such as macrophages and dendritic cells) and secondarily activates NK and T cells, driving anti-tumor immunity through TLR8 agonism. Pembrolizumab (Keytruda) is an established PD-1 immune checkpoint inhibitor that promotes T cell-mediated anti-tumor activity by blocking the PD-1/PD-L1 pathway. The combination leverages SBT6050's innate immune system activation with pembrolizumab's enhancement of adaptive immune cell function, aiming for synergistic anti-tumor effects. The combination is currently under clinical investigation and has demonstrated early tolerability and evidence of immune activation and anti-tumor activity in Phase 1/1b trials in advanced HER2-expressing solid tumors[1][2][3][5].
