This combination consists of SBT6050, a HER2-directed antibody-drug conjugate featuring a Toll-like receptor 8 (TLR8) agonist payload, and trastuzumab deruxtecan, a HER2-directed antibody-drug conjugate with a topoisomerase I inhibitor payload. SBT6050 is designed to activate myeloid cells and indirectly stimulate both innate and adaptive antitumor immunity by targeting HER2-expressing tumor cells via TLR8 agonism. Trastuzumab deruxtecan targets HER2, gets internalized into cells, and releases deruxtecan, a cytotoxic agent that induces DNA damage and apoptosis. This combination is under investigation for the treatment of advanced or metastatic HER2-expressing solid tumors, and leverages two complementary mechanisms targeting the same HER2 antigen—direct cytotoxicity and immune activation[1][3][4][5].
