SBT6290 is an investigational immunotherapeutic agent composed of a monoclonal antibody targeting the cell surface adhesion molecule and tumor-associated antigen nectin-4 (PVRL4), conjugated to a selective Toll-like receptor 8 (TLR8; CD288) agonist. This antibody-drug conjugate is designed for systemic delivery and tumor-localized activation of myeloid cells within the tumor microenvironment. By binding to nectin-4-expressing tumor cells, SBT6290 facilitates targeted delivery of the TLR8 agonist to myeloid cells via Fc gamma receptor engagement, leading to potent activation of proinflammatory cytokine and chemokine production, inflammasome activation, dendritic cell activation, and indirect stimulation of T and NK cell cytolytic activity. Preclinical studies have shown that this approach can drive robust anti-tumor responses in models resistant to checkpoint blockade therapies. The drug is being developed primarily for solid tumors expressing nectin-4, including urothelial carcinoma, triple negative breast cancer (TNBC), non-small cell lung cancer (NSCLC), squamous cell carcinoma of head and neck (SCCHN), hormone receptor positive/HER2-negative breast cancer, and other locally advanced or metastatic solid tumors[1][3][5][6][9].
