SBT8230 is a preclinical **ASGR1-directed TLR8 ImmunoTAC antibody-drug conjugate** originally developed by Silverback Therapeutics for chronic hepatitis B. It couples an anti-ASGR1 monoclonal antibody, intended to localize the conjugate to the liver, with a TLR8-agonist linker-payload. Liver-localized TLR8 activation is designed to activate myeloid cells and promote interferon-gamma-skewed antiviral immune and T-cell responses, with the aim of achieving a functional cure of chronic hepatitis B. Silverback reported preclinical liver enrichment, local myeloid-cell activation, antiviral immune responses, and activity of a mouse surrogate; however, no human clinical study or completed Phase 1 regulatory submission was identified. The program is regarded as discontinued following Silverback's 2022 reverse merger with ARS Pharmaceuticals and the absence of subsequent active development.
