SC-006

SC-006 is an antibody-drug conjugate (ADC) developed by AbbVie for the treatment of advanced colorectal cancer. It consists of a proprietary monoclonal antibody targeting ring finger protein 43 (RNF43), a tumor-associated antigen, linked to a pyrrolobenzodiazepine dimer—a DNA crosslinking cytotoxic agent. The mechanism of action involves both antibody-dependent cell cytotoxicity and direct DNA crosslinking leading to tumor cell death. Clinical development reached phase 1 trials in patients with advanced or metastatic colorectal cancer, including studies as monotherapy and in combination with ABBV-181 (budigalimab). Dose escalation was limited by thrombocytopenia below the predicted efficacious dose, resulting in study termination and discontinuation of further development[1][2][4][7].
