SC-101 is a peptide-drug conjugate (PDC) developed for the treatment of malignant solid tumors that express Nectin-4. The drug consists of a cell-targeting peptide that specifically binds to the human Nectin-4 protein on tumor cells and is conjugated via a protease-hydrolyzable linker to monomethyl auristatin E (MMAE), a microtubule inhibitor. Upon binding to Nectin-4-expressing tumor cells, SC-101 releases its cytotoxic payload, MMAE, which disrupts microtubule function and inhibits tumor growth. The drug was first approved in China in January 2024 for patients with malignant solid tumors expressing Nectin-4 and has received Investigational New Drug (IND) approval from the U.S. FDA for clinical trials in advanced or metastatic solid tumors[1][3][4]. Clinical development includes evaluation as monotherapy and in combination with anti-PD-1 monoclonal antibodies[4].
