SC-CAR.GPC3xIL15.21 is an investigational, autologous "armored" chimeric antigen receptor (CAR) T-cell therapy being developed by Seattle Children's Therapeutics for the treatment of various glypican-3 (GPC3)-expressing solid tumors in both pediatric and adult patients. The T cells are genetically engineered to express a CAR targeting GPC3, a cell-surface protein highly expressed in several pediatric and adult malignancies including hepatocellular carcinoma and various sarcomas. To overcome the challenges of the immunosuppressive tumor microenvironment and improve therapeutic efficacy, the cells are co-engineered to express two stimulatory cytokines, interleukin-15 (IL-15) and interleukin-21 (IL-21), which are intended to promote T-cell survival, expansion, and long-term persistence. Additionally, the product incorporates an inducible caspase 9 (iC9) safety switch, which allows for the rapid elimination of the CAR-T cells upon administration of rimiducid (AP1903) in the event of severe treatment-related toxicities.
