SC-MAGE-A4 is an investigational autologous T-cell receptor (TCR) therapy engineered to target the Melanoma-associated antigen A4 (MAGE-A4) peptide presented by HLA-A*02. Developed by 2seventy bio in collaboration with Regeneron, the therapy incorporates a proprietary TGF-beta "flip" receptor (TGFβRII-41BB). This chimeric receptor is designed to convert the immunosuppressive TGF-beta signal commonly found in the tumor microenvironment into a pro-inflammatory and stimulatory signal via the 4-1BB costimulatory domain. This engineering approach aims to enhance the expansion, persistence, and anti-tumor potency of the engineered T cells against solid tumors. In early 2024, Regeneron acquired 2seventy bio's full oncology pipeline, including the SC-MAGE-A4 program, while JW Therapeutics maintains regional rights for Greater China.
