Overview

SC16.56 is a humanized IgG1 monoclonal antibody that specifically targets delta-like protein 3 (DLL3), a Notch pathway ligand overexpressed in several neuroendocrine tumors, including small cell lung cancer (SCLC) and neuroendocrine prostate cancer (NEPC)[1][2][5]. It was originally developed as the targeting component of an antibody-drug conjugate and has since been used as the basis for radiolabeled diagnostic agents such as [89Zr]Zr-DFO-SC16.56 for immunoPET imaging[1][5][7]. The mechanism of action involves binding to DLL3 on tumor cells, enabling targeted delivery of imaging or therapeutic payloads[2][7]. In clinical studies, radiolabeled forms have demonstrated strong tumor-specific uptake and favorable safety profiles in patients with SCLC and other DLL3-expressing neuroendocrine malignancies[1][5].
