Overview

SC16.8 CAR T cells are a form of chimeric antigen receptor T cell therapy engineered to target delta-like canonical Notch ligand 3 (DLL3), a protein often overexpressed in small cell lung cancer (SCLC) and other neuroendocrine malignancies. The CAR utilizes the SC16.8 antibody clone for DLL3 binding, genetically modifying patient-derived T cells to selectively attack and lyse DLL3-expressing tumor cells. Preclinical studies demonstrated that SC16.8 CAR T cells exhibit significant target-specific cytotoxicity and cytokine (e.g., IFN-γ) secretion upon engagement with DLL3-positive cells. While the basic SC16.8 CAR T cells have shown antitumor activity, their efficacy is enhanced in some settings by co-expression of cytokines such as interleukin-18 (IL-18). The therapy is primarily under investigation for metastatic SCLC and other DLL3-expressing solid tumors[1][2].
