**SC255 is an allogeneic CAR T cell therapy targeting B-cell maturation antigen (BCMA) for the treatment of multiple myeloma.** SC255 uses a BCMA-directed chimeric antigen receptor (CAR) construct licensed from IASO Biotherapeutics and Innovent Biologics, the same construct used in equecabtagene autoleucel (Fucaso; CT103A). The therapy is designed to eliminate malignant plasma cells that express BCMA through immunologic cytotoxicity mediated by allogeneic T lymphocyte replacements. SC255 is engineered with genome modifications to disrupt MHC class I and II, T-cell receptor (TCR) expression (minimizing graft versus host disease), and overexpresses CD47 (enabling evasion of macrophage and natural killer cell-mediated innate immunity). This hypoimmune approach aims to allow the CAR T cells to persist longer in patients without the need for deep immunosuppression, potentially improving treatment durability compared to earlier allogeneic CAR T technologies. The drug aims to be an "off-the-shelf" therapy, providing manufacturing scalability and convenience. SC255 is currently in preclinical development for multiple myeloma[1][2][4].
