SC262 is an investigational allogeneic chimeric antigen receptor (CAR) T cell therapy developed by Sana Biotechnology. It is designed using the company's hypoimmune platform to overcome immunologic rejection of allogeneic cells, potentially resulting in longer CAR T cell persistence and higher rates of durable complete responses in patients with B-cell malignancies. The therapy specifically targets CD22, a protein expressed on the surface of B cells and malignant lymphoma/leukemia cells, including those that have relapsed after or are refractory to prior CD19-directed CAR T therapies. SC262 uses healthy donor-derived CD4+ and CD8+ T cells engineered to express a CAR with a CD22 binder previously tested in academic clinical trials for similar indications. The drug is currently being evaluated in Phase 1 clinical trials for various B-cell malignancies such as non-Hodgkin's lymphoma, diffuse large B-cell lymphoma, follicular lymphoma, mantle cell lymphoma, marginal zone lymphoma, primary mediastinal large B-cell lymphoma (PMBCL), chronic lymphocytic leukemia (CLL), and acute lymphoblastic leukemia (ALL).
