Overview

**SC291** is an investigational allogeneic CAR T-cell therapy developed by Sana Biotechnology using hypoimmune (HIP) technology. It consists of human T lymphocytes from healthy donors, genetically engineered to express a chimeric antigen receptor (CAR) targeting **CD19**, a B-cell surface antigen, with modifications to disrupt MHC class I and II expression and overexpress **CD47** to evade host immune rejection. Administered as a single intravenous dose after lymphodepleting chemotherapy (cyclophosphamide and fludarabine), SC291 enables deeper B-cell depletion for treating relapsed/refractory B-cell malignancies and autoimmune diseases, offering an off-the-shelf alternative to autologous CAR T therapies with scalable manufacturing.
