Scalaradial is a marine-derived sesterterpenoid dialdehyde, originally isolated from sponges such as *Cacospongia mollior*, that serves as a potent pharmacological inhibitor of phospholipase A2 (PLA2) and the TRPM2 (Transient Receptor Potential Melastatin 2) ion channel. As a chanzyme inhibitor, scalaradial blocks TRPM2-mediated calcium influx, which in turn prevents the disruption of the Bip-IRE1α interaction and subsequent inflammatory signaling. In preclinical research, scalaradial has been shown to attenuate aortic inflammation and the progression of thoracic aortic dissection (TAD) by preventing the phenotypic switching of vascular smooth muscle cells (VSMCs) driven by perivascular adipose tissue cytokines. While primarily utilized as a research tool to investigate calcium signaling and inflammatory pathways, its ability to ameliorate vascular degeneration makes it a compound of interest in translational studies for cardiovascular diseases.
