SCD1 short hairpin RNA (shSCD1) is an experimental RNA interference (RNAi) agent designed to silence the expression of the stearoyl-CoA desaturase 1 (SCD1) enzyme. SCD1 is a critical, rate-limiting enzyme that catalyzes the synthesis of monounsaturated fatty acids (MUFAs), such as oleic acid, from saturated fatty acids. In various cancers, including hepatocellular carcinoma (HCC), SCD1 is often upregulated and plays a key role in lipid metabolic reprogramming. This reprogramming increases plasma membrane fluidity, which in turn promotes tumor cell invasive motility and metastasis, particularly in response to high matrix stiffness in the tumor microenvironment. By targeting SCD1 mRNA for degradation, shSCD1 reduces the levels of the SCD1 protein, thereby altering the cellular lipid profile, decreasing membrane fluidity, and inhibiting the metastatic potential of cancer cells. Research has demonstrated that knockdown of SCD1 using shRNA can suppress HCC invasion and metastasis in vivo.
