sCD83 is the soluble form of the human CD83 protein, a member of the immunoglobulin superfamily. Unlike membrane-bound CD83, which is a marker for mature dendritic cells and plays a role in immune activation, sCD83 consists only of the extracellular domain and acts as an immunomodulator with potent immune-suppressive properties. Mechanistically, sCD83 inhibits dendritic cell (DC) maturation and function by disrupting actin cytoskeleton organization via Rab1a/LRRK2 pathways, reducing DC-T cell contact and synapse formation. It induces tolerogenic DCs and regulatory T cells (Tregs), upregulates anti-inflammatory cytokines such as IL-10 and TGF-β, and activates indoleamine 2,3-dioxygenase (IDO), collectively suppressing T cell activation[4][6]. Preclinical studies have shown efficacy in models of autoimmune diseases (e.g., multiple sclerosis/EAE), transplant rejection, wound healing promotion through pro-resolving macrophages induction[8], and more recently in stimulating hair growth by prolonging the anagen phase in hair follicles[8][10]. Mallia Therapeutics is developing topical formulations for androgenetic alopecia with clinical trials planned.
