Overview

scFv-35-DAF is a **recombinant fusion protein** composed of a single-chain variable antibody fragment (scFv-35) that specifically targets the alpha subunit of the acetylcholine receptor (AChR), genetically fused to decay-accelerating factor (DAF), a complement-regulatory protein. The scFv-35 was derived from the monoclonal antibody McAb-35 and provides targeting to the neuromuscular junction (NMJ), while the DAF domain locally inhibits complement activation. This construct was designed for use in myasthenia gravis (MG) as a targeted approach to reduce complement-mediated damage at the NMJ. In preclinical models, scFv-35-DAF demonstrated potent inhibition of complement-mediated lysis and reduced disease severity without blocking neuromuscular transmission or activating complement themselves. The recombinant protein is 529 amino acids (~57.7 kDa), produced using recombinant technologies[1].
