scFv BCMA CAR T cells represent a target-specific platform of autologous chimeric antigen receptor (CAR) T-cell therapies engineered to express a CAR containing a single-chain variable fragment (scFv) specific for B-cell maturation antigen (BCMA). BCMA is a cell surface protein highly and selectively expressed on malignant plasma cells in multiple myeloma. The CAR construct typically consists of an extracellular scFv binding domain, a transmembrane domain, and intracellular signaling domains, most commonly including a co-stimulatory domain (such as 4-1BB/CD137 or CD28) and an activating CD3-zeta domain. Upon infusion, these engineered T cells recognize BCMA-expressing tumor cells, leading to T-cell activation, expansion, and targeted cytotoxicity. This platform technology underpins several approved and investigational therapies, including idecabtagene vicleucel and ciltacabtagene autoleucel, and is a cornerstone of the current treatment landscape for relapsed or refractory multiple myeloma.
