scFv-Fc C21 is a fully human monoclonal antibody fragment targeting chondroitin sulfate proteoglycan 4 (CSPG4), a cell surface protein highly expressed in melanoma and other cancers. Developed by researchers at the University of Pittsburgh Cancer Institute, the molecule is a fusion protein consisting of a single-chain variable fragment (scFv) isolated from a phage display library and a human IgG1 Fc region. scFv-Fc C21 binds specifically to CSPG4, inhibiting signaling pathways critical for tumor cell survival, growth, and motility, including the activation of FAK, Erk1/2, PDK1, and Akt. Preclinical studies have demonstrated that scFv-Fc C21 can significantly inhibit melanoma cell migration and growth in vitro and prevent post-surgical tumor recurrence and lung metastasis in vivo.
