Overview

scFv47 is a therapeutic single-chain variable fragment (scFv) that targets the Interleukin-13 receptor alpha 2 (IL13Rα2), a cell surface receptor significantly overexpressed in glioblastoma multiforme. It acts as a competitive antagonist, binding to the IL13Rα2 receptor to prevent its interaction with the natural ligand IL13. Structural studies indicate that the scFv47 heavy chain complementarity-determining regions (CDRs) engage the D2 domain of the receptor, while the light chain CDR1 primarily contacts the D1 domain. With a measured binding affinity (KD) of approximately 1.39 nM, scFv47 is being developed as a targeting domain for Chimeric Antigen Receptor (CAR) T-cell therapies and as a template for rational antibody engineering to treat high-grade gliomas.
