Overview

scFv:TRAIL-armed autologous T cells is an experimental, preclinical cell-based immunotherapy being developed by the University Medical Center Groningen (Universitair Medisch Centrum Groningen) for the treatment of melanoma. This therapeutic approach involves isolating autologous T cells from surgical melanoma patients and ex vivo "arming" them with a recombinant fusion protein consisting of a single-chain variable fragment (scFv) antibody linked to Tumor Necrosis Factor-Related Apoptosis-Inducing Ligand (TRAIL). The scFv component (typically targeting CD3 or CD7) serves to anchor the fusion protein to the T-cell surface, thereby decorating the T cells with high levels of active TRAIL. Upon administration, these armed T cells target melanoma cells, where the surface-bound TRAIL interacts with death receptors TRAIL-R1 (DR4) and TRAIL-R2 (DR5) to trigger apoptotic signaling and tumor cell death. This platform aims to enhance the intrinsic tumoricidal activity of adoptive T-cell therapies.
