SCG142 is a next-generation, autologous, human papillomavirus (HPV)-specific T-cell receptor-engineered T cell (TCR-T) therapy developed by SCG Cell Therapy. It is designed for the treatment of advanced or metastatic HPV-associated solid tumors, including cervical cancer, head and neck cancer, anal cancer, vulvar cancer, penile cancer, and vaginal cancer. The therapy utilizes high-avidity natural TCRs that specifically recognize the E7 oncoprotein from HPV16 and HPV52 genotypes presented by HLA-A*02:01. These engineered T cells are further "armored" with a chimeric switch receptor composed of transforming growth factor beta receptor type 2 (TGFβRII) fused to tumor necrosis factor receptor superfamily member 9 (4-1BB), which converts immunosuppressive signals in the tumor microenvironment into co-stimulatory signals to enhance anti-tumor activity and persistence. Preclinical studies have shown that SCG142 exhibits robust CD8+ and CD4+ proliferation and tumor inhibition both in vitro and in vivo without alloreactivity or off-target toxicity[3][6][7][8].
