SCH-900271 is a potent, orally active small molecule agonist of the nicotinic acid receptor, also known as hydroxycarboxylic acid receptor 2 (HCAR2 or GPR109A). It was developed for the treatment of dyslipidemia, including mixed hyperlipidemia and primary hypercholesterolemia. The drug demonstrated strong activity in preclinical models by significantly reducing plasma free fatty acids and improving lipid profiles without causing significant flushing—a common side effect of traditional niacin therapies. SCH-900271 advanced to phase II clinical trials but its development has since been discontinued[1][2][3][4][5][6].
