Overview

Schisantherin B is a natural lignan compound primarily isolated from plants of the genus Schisandra, including Schisandra chinensis and Schisandra sphenanthera. It is known for a range of biological activities, including neuroprotective, antidepressant, anti-inflammatory, anti-asthmatic, and anticancer effects. Mechanistically, schisantherin B modulates several molecular pathways, notably the NF-κB signaling pathway (involved in inflammation), peroxisome proliferator-activated receptors (PPAR), and interleukin 17 (IL-17) signaling. It has been shown to reduce the formation of advanced glycation end products (AGEs), inhibit pro-inflammatory gene expression, suppress oxidative stress, modulate neurotransmitter levels in the brain, inhibit cancer cell growth via apoptosis induction, and protect against neurodegenerative changes, including tau phosphorylation and neuroinflammation in preclinical studies. In animal models, schisantherin B is distributed widely, particularly accumulating in the liver and kidneys, and can cross the blood-brain barrier, highlighting potential for CNS indications. These diverse pharmacological effects make schisantherin B a promising candidate for the treatment of neurodegenerative diseases, depression, asthma, and certain cancers[1][4][6][7][8][9][10].
