SchistoShield is a recombinant protein subunit vaccine candidate designed for the prevention of schistosomiasis, specifically targeting *Schistosoma mansoni*. The vaccine is composed of the Sm-p80 antigen, which is the large subunit of the parasite's calcium-activated neutral protease (calpain), formulated with the GLA-SE adjuvant (a synthetic Toll-like receptor 4 agonist in a stable oil-in-water emulsion). Sm-p80 is a critical protein for the parasite's tegumental membrane renewal and survival across various life stages. Developed through a partnership between PAI Life Sciences, Texas Tech University Health Sciences Center, and the Leiden University Medical Center (LUMC), SchistoShield aims to induce high titers of functional antibodies and Th1-biased cellular immune responses to reduce worm burden and egg production. It is currently being evaluated in Phase 1 and Phase 2 clinical trials, including controlled human infection models (CHIM) to assess safety and preliminary efficacy.
