Overview

Schweinfurthin-3114 (S-3114) is a natural product-derived small molecule belonging to the schweinfurthin family, originally purified from plant extracts. It is currently under investigation as a potential therapeutic for multiple myeloma. S-3114 acts by inhibiting the Phosphoinositide 3-Kinase (PI3K)/Akt signaling pathway, which leads to cell cycle arrest at the G2/M phase and the induction of apoptosis through the extrinsic pathway, specifically involving the cleavage of caspase 8. Research also indicates that S-3114 disrupts Golgi-mediated secretion, impacting the production and release of Macrophage Inflammatory Protein-1 alpha (MIP-1α), a chemokine critical for the progression of malignant plasma cell diseases.
