SCIB1 is a genetically engineered DNA cancer vaccine developed by Scancell for the treatment of melanoma. It consists of a plasmid DNA encoding a human IgG1 antibody molecule that has been engineered to express cytotoxic T cell epitopes from the melanoma antigens Tyrosinase-Related Protein 2 (TRP2) and gp100, as well as helper T cell epitopes[3][6][8]. Upon intramuscular administration, the plasmid is taken up and expressed in host cells, leading to secretion of an engineered antibody that targets dendritic cells via the high-affinity Fc receptor CD64. This results in both direct presentation and cross-presentation of tumor-associated epitopes on MHC class I and II molecules, stimulating robust CD4+ and CD8+ T cell responses against melanoma cells[6][7][8]. The dual mechanism enhances both frequency and avidity of anti-tumor T cells. SCIB1 has shown promising activity in combination with immune checkpoint inhibitors such as nivolumab and ipilimumab in advanced melanoma clinical trials[5].
