A **combination immunotherapy regimen** consisting of the DNA plasmid cancer vaccine SCIB1, administered together with the immune checkpoint inhibitors nivolumab and ipilimumab. SCIB1 encodes epitopes from tumor antigens TRP-2 and gp100, designed within an antibody framework to target activated dendritic cells and stimulate robust CD4 and CD8 T-cell responses. Nivolumab is a human monoclonal antibody targeting programmed cell death protein 1 (PD-1), while ipilimumab is a human monoclonal antibody targeting cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4). The combination aims to synergistically enhance anti-tumor immunity in advanced, unresectable melanoma patients, particularly those who may not optimally respond to checkpoint inhibitor monotherapy. Clinical evaluation in the phase 2 SCOPE trial has shown promising objective response rates and disease control.
