**SCIB2** is a DNA-based cancer vaccine from Scancell's ImmunoBody platform, designed as a fusion protein encoding a human IgG1 antibody with 16 NY-ESO-1 epitopes incorporated into its CDR regions to target over 80% of HLA phenotypes. It enhances uptake and presentation of the NY-ESO-1 cancer-testis antigen by antigen-presenting cells, inducing high-frequency, high-avidity CD4+ and CD8+ T cell responses that demonstrate potent cytotoxicity against NY-ESO-1-expressing tumor cells. Preclinical studies in mice showed SCIB2 monotherapy achieving 35% long-term survival in established B16-NY-ESO-1 tumors, with enhanced efficacy (up to 100% survival) in combinations with Treg depletion, CTLA-4 blockade, or PD-1 blockade; it outperforms peptide vaccines in T cell avidity and tumor control. Developed by Scancell for solid tumors like NSCLC, melanoma, ovarian, and prostate cancers, with planned nanoparticle delivery in Phase 1/2 trials.[1][2][3][4]
