scIL-12 nanogel is a cytokine-loaded protein nanoparticle designed as a synthetic "backpack" for adoptive cell therapy (ACT). Developed at the Koch Institute for Integrative Cancer Research at MIT, it consists of single-chain interleukin-12 (scIL-12) crosslinked with disulfide-containing linkers to form a nanogel. These nanogels are chemically conjugated to the surface of tumor-specific T cells. The release of scIL-12 is triggered by T cell receptor (TCR) signaling, which induces changes in the cell surface redox state, allowing for localized cytokine delivery within the tumor microenvironment. This approach aims to enhance the efficacy of ACT in solid tumors, such as hepatocellular carcinoma and melanoma, while minimizing the systemic toxicity associated with free IL-12 administration.
