Overview

SCIO-469 is a selective small molecule inhibitor of p38α mitogen-activated protein kinase (MAPK). Developed by Scios (a subsidiary of Johnson & Johnson), it was designed to modulate inflammatory responses by inhibiting the production of cytokines such as TNF-α and IL-6. The compound exhibits high potency for the p38α isoform (MAPK14) with approximately 10-fold selectivity over p38β and minimal activity against other kinases. SCIO-469 was advanced into Phase 2 clinical trials for several indications, including myelodysplastic syndrome (MDS), rheumatoid arthritis, and multiple myeloma. Despite showing biological activity in reducing inflammatory markers, clinical development was ultimately discontinued for these indications.
