Drug intelligence / Profile preview
scL-siHER2 (SGT-78) is an investigational RNA interference (RNAi) therapeutic designed to target cancers overexpressing the Human Epidermal Growth Factor Receptor 2 (HER2). It utilizes a proprietary tumor-targeting nanodelivery system known as **scL**, which consists of a cationic liposome decorated with a single-chain antibody fragment (scFv) targeting the transferrin receptor (TfR). This allows the nanoparticle to specifically bind to and be internalized by cancer cells, which often overexpress TfR. The payload is a small interfering RNA (siRNA) molecule (sometimes a DNA-RNA hybrid) that is complementary to HER2 mRNA. Once inside the cytoplasm, the siRNA triggers the RNA-induced silencing complex (RISC) to degrade the HER2 mRNA, preventing the translation of the HER2 protein. This gene silencing inhibits the signaling pathways responsible for tumor cell proliferation and survival, and has been shown to sensitize resistant tumors to chemotherapy in preclinical models of pancreatic, breast, and ovarian cancers.
