Drug intelligence / Profile preview
A sclerostin-neutralizing antibody (Scl-Ab) is a therapeutic monoclonal antibody designed to target and inhibit sclerostin, a glycoprotein encoded by the *SOST* gene and primarily secreted by osteocytes. Sclerostin acts as a negative regulator of bone formation by binding to low-density lipoprotein receptor-related protein 5/6 (LRP5/6) co-receptors, thereby antagonizing the canonical Wnt/β-catenin signaling pathway. By neutralizing sclerostin, these antibodies prevent its inhibitory interaction with LRP5/6, restoring Wnt signaling and promoting osteoblast differentiation, proliferation, and bone-forming activity (osteoanabolism) while simultaneously reducing bone resorption. Sclerostin-neutralizing antibodies, such as the FDA-approved romosozumab, are primarily developed for the treatment of postmenopausal osteoporosis and other bone-loss disorders, though they are also investigated in preclinical models for conditions like periodontitis, osteogenesis imperfecta, and bone fracture healing.
