SCN9A-targeting spherical nucleic acid is a preclinical non-opioid analgesic developed by Exicure, Inc. using its proprietary Spherical Nucleic Acid (SNA) technology platform. The therapeutic consists of a nanoparticle core (typically gold or lipid-based) functionalized with a dense, highly oriented shell of antisense oligonucleotides. This unique three-dimensional structure allows for enhanced cellular uptake and resistance to nuclease degradation without the need for additional transfection agents. The drug is designed to selectively downregulate the expression of the SCN9A gene, which encodes the voltage-gated sodium channel Nav1.7. Nav1.7 is a genetically validated target for pain management, as it is primarily expressed in peripheral sensory neurons and plays a critical role in the initiation and conduction of pain signals. By reducing Nav1.7 channel density, the SNA candidate aims to provide potent and long-lasting pain relief. However, Exicure discontinued the development of this program in late 2021 as part of a strategic restructuring to focus on other pipeline priorities.
