SCO-101 + FOLFIRI is an investigational drug combination comprised of SCO-101, a small molecule inhibitor of ABCG2, UGT1A1, and SRPK1, administered together with the FOLFIRI chemotherapy regimen (folinic acid [leucovorin], fluorouracil [5-FU], irinotecan) for treatment of metastatic colorectal cancer (mCRC). SCO-101 modulates resistance mechanisms and increases exposure of irinotecan's active metabolite SN-38, potentially improving efficacy in patients who have progressed on standard therapies. In multiple parts of the CORIST phase II trial, SCO-101 was given orally for several days (typically days 1–6) and FOLFIRI intravenously (days 5–7) in recurring two-week cycles. This combination has shown promising activity, increasing progression-free survival in a drug-resistant mCRC population and maintaining a favorable safety profile[1][3][5].
