Drug intelligence / Profile preview
SCP-lenalidomide is a preclinical small molecule candidate being developed by Nexus Alliance Biopharmaceuticals for the treatment of glioblastoma. The drug utilizes the company's proprietary Self-Correcting Polymer (SCP) technology to formulate lenalidomide, an established immunomodulatory drug (IMiD). Lenalidomide acts as a molecular glue that binds to cereblon (CRBN), a component of the CRL4-CRBN E3 ubiquitin ligase complex. This binding event modifies the substrate specificity of the ligase, leading to the recruitment, ubiquitination, and subsequent proteasomal degradation of specific transcription factors, most notably Ikaros (IKZF1) and Aiolos (IKZF3). In glioblastoma, this SCP-formulated version is intended to optimize the delivery and therapeutic efficacy of lenalidomide, potentially enhancing its anti-tumor and anti-angiogenic properties within the central nervous system.
