Overview

ScPvs25 is an experimental recombinant protein transmission-blocking vaccine (TBV) developed by the National Institute of Allergy and Infectious Diseases (NIAID) to prevent the transmission of *Plasmodium vivax* malaria. The vaccine consists of the Pvs25 protein, which is expressed on the surface of the malaria parasite during its zygote and ookinete stages within the mosquito midgut. The protein is produced using a *Saccharomyces cerevisiae* (yeast) expression system. ScPvs25 is designed to induce the production of specific antibodies in the human host. When a mosquito bites a vaccinated individual and ingests these antibodies, they bind to the Pvs25 protein on the parasite's surface, inhibiting its development within the mosquito and preventing the mosquito from transmitting the disease to other humans. In clinical trials, ScPvs25 has been formulated with the adjuvant Montanide ISA 51 to enhance immunogenicity.
