Overview

SCR-8388 is a potent and selective small molecule inhibitor of the Son of Sevenless 1 (SOS1) protein, developed by Simcere Pharmaceuticals. It acts by blocking the interaction between SOS1 and KRAS-GDP, which prevents the exchange of GDP for GTP and thereby inhibits KRAS activation. SCR-8388 has demonstrated broad-spectrum activity against various KRAS mutations, including G12D, G12C, G12S, and G13D, in preclinical models. It is being investigated for the treatment of KRAS-addicted cancers, both as a monotherapy and in combination with other agents such as EGFR antagonists.
